| IDT offers qPCR positive controls for each of the H1N1 primer/probe sets as well as a universal plasmid control containing all four cloned amplicons. | |
| The qPCR primer/probe set controls are ReadyMadeTM DNA Ultramers that can be used as copy number standards for each assay. Each Ultramer control contains an additional 14 bases (shown as lower case bases in Figure 1). By increasing the size of the amplicon, it is possible to distinguish a positive result as coming from the control or as a possible viral contaminant by running the product on a gel. Finally, each Ultramer Control also contains SP6 and T7 promoter regions. The promoter regions allow for the generation of in vitro transcribed RNA if an RNA transcript control is desired. Each tube contains 6x1013 molecules. | |

| The universal plasmid control contains positive control sequences for all four target sequences in the swine flu testing panel. The plasmid also contains restriction sites for digestion which will allow for degradation if amplicon contamination occurs. Similar to the Ultramer controls, the plasmid also contains SP6 and T7 promoter regions for generation of in vitro transcribed RNA. The design of the plasmid can be seen in Figure 2. | |
