All oligos synthesized at IDT are analyzed by mass spectrometry. Matrix-Assisted Laser Desorption/Ionization-Time Of Flight (MALDI-TOF) mass spectrometry is suitable for use in a high-throughput QC setting and can resolve oligonucleotides up to 50 bases in length (~15,000 Daltons). Electrospray Ionization (ESI) mass spectrometry can resolve longer oligonucleotides (up to ~150 bases in length).


CE provides quantitative analysis of micropurity. Nanoliter samples of oligonucleotides are electrokinetically injected into a fused silica capillary and filled with a denaturing gel under an electric potential. The oligonucleotides move through the gel matrix and are separated according to length. Absorbance at 260nm is detected as the oligo passes through a window in the capillary. The resulting electropherogram provides the percent purity of the full-length product as well as truncated products.


HPLC is a universally accepted method of sample quality assessment. IDT employees use both Reverse-Phase and Ion-Exchange methods to assess product purity. These methods are employed on purified oligos where the composition is incompatible with CE technology.
